    A diversity of dusty AGN tori: Data release for the VLTI/MIDI AGN Large Program and first results for 23 galaxies

    The AGN-heated dust distribution (the "torus") is increasingly recognized not only as the absorber required in unifying models, but as a tracer for the reservoir that feeds the nuclear Super-Massive Black Hole. Yet, even its most basic structural properties (such as its extent, geometry and elongation) are unknown for all but a few archetypal objects. Since most AGNs are unresolved in the mid-infrared, we utilize the MID-infrared interferometric Instrument (MIDI) at the Very Large Telescope Interferometer (VLTI) that is sensitive to structures as small as a few milli-arcseconds (mas). We present here an extensive amount of new interferometric observations from the MIDI AGN Large Program (2009 - 2011) and add data from the archive to give a complete view of the existing MIDI observations of AGNs. Additionally, we have obtained high-quality mid-infrared spectra from VLT/VISIR. We present correlated and total flux spectra for 23 AGNs and derive flux and size estimates at 12 micron using simple axisymmetric geometrical models. Perhaps the most surprising result is the relatively high level of unresolved flux and its large scatter: The median "point source fraction" is 70 % for type 1 and 47 % for type 2 AGNs meaning that a large part of the flux is concentrated on scales smaller than about 5 mas (0.1 - 10 pc). Among sources observed with similar spatial resolution, it varies from 20 % - 100 %. For 18 of the sources, two nuclear components can be distinguished in the radial fits. While these models provide good fits to all but the brightest sources, significant elongations are detected in eight sources. The half-light radii of the fainter sources are smaller than expected from the size ~ L^0.5 scaling of the bright sources and show a large scatter, especially when compared to the relatively tight size--luminosity relation in the near-infrared.Comment: A&A in press; 93 pages, 63 figures, 39 tables; data available only via CD

    The dusty heart of Circinus II. Scrutinizing the LM-band dust morphology using MATISSE

    In this paper we present the first-ever LL- and MM-band interferometric observations of Circinus, building upon a recent NN-band analysis. We used these observations to reconstruct images and fit Gaussian models to the LL and MM bands. Our findings reveal a thin edge-on disk whose width is marginally resolved and is the spectral continuation of the disk imaged in the NN band to shorter wavelengths. Additionally, we find a point-like source in the LL and MM bands that, based on the LMNLMN-band spectral energy distribution fit, corresponds to the NN-band point source. We also demonstrate that there is no trace of direct sightlines to hot dust surfaces in the circumnuclear dust structure of Circinus. By assuming the dust is present, we find that obscuration of AV250_{\rm V} \gtrsim 250 mag is necessary to reproduce the measured fluxes. Hence, the imaged disk could play the role of the obscuring "torus" in the unified scheme of active galactic nuclei. Furthermore, we explored the parameter space of the disk + hyperbolic cone radiative transfer models and identify a simple modification at the base of the cone. Adding a cluster of clumps just above the disk and inside the base of the hyperbolic cone provides a much better match to the observed temperature distribution in the central aperture. This aligns well with the radiation-driven fountain models that have recently emerged. Only the unique combination of sensitivity and spatial resolution of the VLTI allows such models to be scrutinized and constrained in detail. We plan to test the applicability of this detailed dust structure to other MATISSE-observed active galactic nuclei in the future.Comment: Main article: 12 pages, 6 figures.     Recent translational research: circulating tumor cells in breast cancer patients

    In breast cancer patients, hematogenous tumor cell dissemination can be detected, even at the single cell level, by applying immunocytochemical and molecular assays. Various methods for the detection of circulating tumor cells in the peripheral blood have been described. Results from recently reported studies suggest that circulating tumor cell levels may serve as a prognostic marker and for the early assessment of therapeutic response in patients with metastatic breast cancer. However, in early-stage breast cancer, the impact of circulating tumor cells is less well established than the presence of disseminated tumor cells in bone marrow; several clinical studies have demonstrated that cells of the latter type are an independent prognostic factor at primary diagnosis. In this article we briefly summarize recent studies examining the presence of circulating tumor cells in the blood and discuss further clinical applications

    There is an increasing public interest in sea ice information from both Polar Regions, which requires up-to-date background information and data sets at different levels for various target groups. In order to serve this interest and need, seaiceportal.de (originally: meereisportal.de) was developed as a comprehensive German knowledge platform on sea ice and its snow cover in the Arctic and Antarctic. It was launched in April 2013. Since then, the content and selection of data sets increased and the data portal received increasing attention, also from the international science community. Meanwhile, we are providing near-real time and archive data of many key parameters of sea ice and its snow cover. The data sets result from measurements acquired by various platforms as well as numerical simulations. Satellite observations of sea ice concentration, freeboard, thickness and drift are available as gridded data sets. Sea ice and snow temperatures and thickness as well as atmospheric parameters are available from autonomous platforms (buoys). Additional ship observations, ice station measurements, and mooring time series are compiled as data collections over the last decade. In parallel, we are continuously extending our meta-data and uncertainty information for all data sets. In addition to the data portal, seaiceportal.de provides general comprehensive background information on sea ice and snow as well as expert statements on recent observations and developments. This content is mostly in German in order to complement the various existing international sites for the German speaking public. We will present the portal, its content and function, but we are also asking for direct user feedback

    Targeting cyclin B1 inhibits proliferation and sensitizes breast cancer cells to taxol

    Background Cyclin B1, the regulatory subunit of cyclin-dependent kinase 1 (Cdk1), is essential for the transition from G2 phase to mitosis. Cyclin B1 is very often found to be overexpressed in primary breast and cervical cancer cells as well as in cancer cell lines. Its expression is correlated with the malignancy of gynecological cancers. Methods In order to explore cyclin B1 as a potential target for gynecological cancer therapy, we studied the effect of small interfering RNA (siRNA) on different gynecological cancer cell lines by monitoring their proliferation rate, cell cycle profile, protein expression and activity, apoptosis induction and colony formation. Tumor formation in vivo was examined using mouse xenograft models. Results Downregulation of cyclin B1 inhibited proliferation of several breast and cervical cancer cell lines including MCF-7, BT-474, SK-BR-3, MDA-MB-231 and HeLa. After combining cyclin B1 siRNA with taxol, we observed an increased apoptotic rate accompanied by an enhanced antiproliferative effect in breast cancer cells. Furthermore, control HeLa cells were progressively growing, whereas the tumor growth of HeLa cells pre-treated with cyclin B1 siRNA was strongly inhibited in nude mice, indicating that cyclin B1 is indispensable for tumor growth in vivo. Conclusion Our data support the notion of cyclin B1 being essential for survival and proliferation of gynecological cancer cells. Concordantly, knockdown of cyclin B1 inhibits proliferation in vitro as well as in vivo. Moreover, targeting cyclin B1 sensitizes breast cancer cells to taxol, suggesting that specific cyclin B1 targeting is an attractive strategy for the combination with conventionally used agents in gynecological cancer therapy

    Dvl2-Dependent Activation of Daam1 and RhoA Regulates Wnt5a-Induced Breast Cancer Cell Migration

    The Dishevelled (Dvl) and Dishevelled-associated activator of morphogenesis 1 (Daam1) pathway triggered by Wnt5a regulates cellular polarity during development and tissue homoeostasis. However, Wnt5a signaling in breast cancer progression remains poorly defined.We showed here that Wnt5a activated Dvl2, Daam1 and RhoA, and promoted migration of breast cancer cells, which was, however, abolished by Secreted Frizzled-related protein 2 (sFRP2) pretreatment. Dominant negative Dvl2 mutants or Dvl2 siRNA significantly decreased Wnt5a-induced Daam1/RhoA activation and cell migration. Ectopic expression of N-Daam1, a dominant negative mutant, or Daam1 siRNA remarkably inhibited Wnt5a-induced RhoA activation, stress fiber formation and cell migration. Ectopic expression of dominant negative RhoA (N19) or C3 exoenzyme transferase, a Rho inhibitor, decreased Wnt5a-induced stress fiber formation and cell migration.Taken together, we demonstrated for the first time that Wnt5a promotes breast cancer cell migration via Dvl2/Daam1/RhoA

    TRY plant trait database – enhanced coverage and open access

    Plant traits - the morphological, anatomical, physiological, biochemical and phenological characteristics of plants - determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait‐based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits - almost complete coverage for ‘plant growth form’. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait–environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives
